Description

Oligonucleotides are synthetically produced biomolecules that have proven to be crucial for gene editing therapies. Biotech companies put a lot of time and effort in optimizing and developing groundbreaking analysis techniques for these molecules; purity assessment being one of the most significant ones. High performance liquid chromatography (HPLC), is one of the tools almost every research team uses to assess and characterize the constituents found in a compound. Knowing this technique becomes a very important skill even for entry-level jobs.

In this Build Project, you'll wear the hat of an Analytical Development Scientist and help optimize a method for purity/integrity assessment of oligonucleotides through RP-IP HPLC. Under the supervision of an experienced Build Fellow, you'll become proficient in navigating scientific literature and building your own conclusions and course of actions in the development of this method. You'll learn the science behind liquid chromatography and how biomolecules such as RNA behaves in such. You will also be able to appreciate role that oligonucleotides play in gene editing therapies; a rising focus for biotech companies. All this will happen in an environment that simulates the operations of a real Analytical Development team.

  • Use research papers to develop and optimize analytical methods.
  • Learn how to apply organic chemistry concenpts to real life scenarios in order to better characterize biomolecules.
  • Learn how to analyze and interpret HPLC data.
  • Understand how HPLC instruments function, including their different modalities and uses.
Build Projects are 8-week experiences that operate on a rolling basis. Selected participants engage in weekly live workshops with a Build Fellow and 2-15 other students.

Prerequisites

  • Basic knowledge of biochemistry: you should understand the function of DNA in cells and how RNA molecules work in our body.
  • Entry-level knowledge of organic chemistry: you shouldhave basic understanding of  the structure, properties, and reactions of organic compounds such as alcohols, amines and alkyls.
  • Basic/intermediate knowledge of analytical chemistry and instrumental analysis: you should be able to understand how separation techniques such as chromatography are used to analyze compounds and materials.
  • Other prerequisites: before the project starts, you'll need to have google scholarly and Microsoft Word/PowerPoint installed on your computer.
